After skipper KL Rahul won the toss and opted to bowl, India restricted the visitors to 270, powered by superb spells from Prasidh Krishna and Kuldeep Yadav, who picked four wickets each.
Quinton de Kock was the lone standout for South Africa, scoring a fighting century.
In reply, India’s chase was dominated by a majestic opening stand of 155 between Rohit Sharma and Yashasvi Jaiswal.
Rohit struck 75, while Jaiswal continued in sublime touch, registering his maiden ODI hundred and remaining unbeaten on 116. He later added a century partnership with Virat Kohli, who finished on 65*, his fourth consecutive 50-plus ODI score.
India cruised to 271/1 in 39.5 overs, sealing an emphatic series victory.
